Col-0 is resistant to the isolate AG8 and susceptible to the isolate AG2-1 of soil-borne necrotrophic fungus Rhizoctonia solani. Freezing tolerance of non-acclimated Col-0 leaves: LT50 (temperature of 50% electrolyte leakage) = −5.34°C; freezing tolerance of Col-0 leaves after acclimation at 4°C under 16h photoperiod (90µmol photons m-2 s-1) for 14 days: LT50 = −9.68°C. Knockout mutant carrying T-DNA insertion (SALK_104713) in the intron of TBF1 (TL1-binding factor; At4g36990). TBF1 is a heat-shock factor-like transcription factor that specifically binds to the TL1 (GAAGAAGAA) cis element enriched in promoters of genes regulated by microbe-associated molecular patterns (e.g. EF-Tu(elf26), flg22) and in promoters of some salicylic acid (SA)-induced genes encoding endoplasmic reticulum-localized proteins. In the presence of 150-250µM SA or 10µM EF-Tu(elf18), growth of tbf1 seedlings is less inhibited than growth of Col-0 wild type. Rosette stage wild type plants pre-treated with SA or EF-Tu(elf18) become more resistant to bacterial pathogen Pseudomonas syringae pv. maculicola ES4326 whereas tbf1 plants pre-treated the same way remain almost as susceptible to the pathogen as the mock-treated tbf1 and wild type plants. Flg22-induced resistance to P. syringae pv. maculicola ES4326 is not compromised in tbf1. Compared to the wild type, tbf1 is more susceptible to tunicamycin, an inducer of unfolded protein response (Pajerowska-Mukhtar et al., 2012, Curr Biol. 22: 103-112).
