Xenograft-derived MCF-7 cells isolated from primary tumor of breast in non-ovarioectomized female NCR/Nu/Nu nude mice. Four-week-old non-ovarioectomized female NCR/Nu/Nu nude mice were anesthetized by exposure to 3% isoflurane and injected into mammary fat pad with cells (MCF-7 cell line transfected with empty retroviral vector encoding neomycine resistance gene) suspended in 50 ul of 50% Matrigel. After 12 weeks, mice were sacrificed and primary xenograft tumors were processed. Tumor cells were isolated from primary tumor and expanded in culture for further use. It is not further specified whether the microarray analysis was performed on freshly isolated tumor cells or in vitro expanded cells.
