Side population (SP) subset of malignant fibrous histiocytoma tumor cells (MFH). Cells were divided into the SP group based on ability to efflux Hoechst dye. Cells were treated either alone with 2.5 μg/mL of Hoechst 33342 dye or in combination with 50 μmol/L verapamil, an inhibitor of ABC transporters. Cells were counterstained with 1 μg/mL of propidium iodide (PI), and PI-positive nonviable cells were excluded. To detect for SP, cells were analyzed by using a dual wavelength analysis (blue, 424–444 nm; red, 675 nm) after excitation with 350 nm UV light. During flow cytometry analysis, negatively stained cells fall to the “side-population” subset. Malignant fibrous histiocytoma tumor tissue (also termed as undifferentiated pleomorphic sarcoma) was obtained at the time of the initial biopsy. Pathology was verified by an independent experienced musculoskeletal pathologist, and for the absence of staining for routine markers used to identify sarcoma subtypes. The samples were mechanically dissociated and then digested. | |
