Mature IL-4 dendritic cells differentiated from CD14+ monocytes obtained from healthy donors. Monocytes were seeded in RPMI and supplemented with 2.5 % heat-inactivated human AB serum. Monocytes were cultured with 800 IU/mL granulocyte macrophage colony-stimulating factor and 20 ng/mL IL-4 in order to generate immature IL-4 DCs. Conventional maturation cocktail, comprising 10 ng/mL tumor necrosis factor-α, 10 ng/mL interleukin-1β, 15 ng/ml interleukin-6 and 1 µg/mL prostaglandin E2 was added for 48 hours.
