Mature IL-4 dendritic cells differentiated from CD14+ monocytes obtained from healthy donors. Monocytes were seeded in RPMI and supplemented with 2.5 % heat-inactivated human AB serum. Monocytes were cultured with 800 IU/mL granulocyte macrophage colony-stimulating factor and 20 ng/mL IL-4 in order to generate immature IL-4 DCs. Peripheral blood mononuclear cells stimulated for 9 hours with 40% plasma derived from gram-positive pneumonia patients. Bacterial infection was detected in every sample and all patients showed acute illness, a pulmonary infiltrate or consolidation (detected by a radiologist) and at least one of the following symptoms: Body temperature > 37.8 °C or < 36°C, peripheral white blood cell number > 10 000 ul or < 4500 ul or >15% immature neutrophils. Furthermore patients also revealed two of the following three factors: tachypnea, dyspnea or hypoxemia. Samples were collected within the first day of hospitalization.
Control healthy plasma treated PBMC sample
