Motor neurons from 15 weeks old mice of B6.Cg-Tg(SOD1-G93A)1Gur/J mouse strain have been sampled by laser microdissection. About 300 motor neurons were sampled per mouse (n = 7). For the laser microdissections 8 um coronal section of lumbar spinal cord was prepared and Leica Laser Microdissection system was used. These mice were treated with vehicle (250 mM sorbitol, 0.004% Tween-80 and 10 mM sodium acetate buffer). At this age mice presented clear motor impairment and motor neuron degeneration is documented.
Control amyotrophic lateral sclerosis study 2 (11 weeks)
Motor neurons from 11 weeks old mice of B6.Cg-Tg(SOD1-G93A)1Gur/J mouse strain have been sampled by laser microdissection. About 300 motor neurons were sampled per mouse (n = 8). For the laser microdissections 8 um coronal section of lumbar spinal cord was prepared and Leica Laser Microdissection system was used. At this age mice present no signs of motor dysfunction but subtle signs of denervation detectable by electromyography.
