Experimental RhoA knockout study 1 (in vivo)
Freshly isolated RhoA-null keratinocytes obtained from epidermis of RhoA (fl/fl) K5 Cre mice. Skin samples were washed for 5 min in PBS containing antibiotics followed by digestion with 0.8% trypsin in PBS (50 min; 37 °C). Obtained epidermis was shaken for 30 min in low-calcium MEM supplemented with 0.25 mg/ml DNAase I and 8 % FCS treated with Chelex. Cell suspension was filtered (70 um) and collected keratinocytes were washed once with DNase I medium, resuspended and analyzed. RhoA: Ras homolog gene family, member A.
Control RhoA knockout study 1 (in vitro)
Cultured primary RhoA-null keratinocytes obtained from epidermis of RhoA (fl/fl) K5 Cre mice. Primary keratinocytes were cultured in low-calcium MEM containing 5 ug/ml insulin, 10 ng/ml epidermal growth factor, 10 ug/ml transferrin, 10 uM phosphoethanolamine, 10 uM ethanolamine, 0.36 ug/ml hydrocortisone, 2 mM L-glutamine, 100 U/ml penicillin, 100 ug/ml streptomycin and 8% Chelex-treated FCS. Motor neurons from 15 weeks old mice of B6.Cg-Tg(SOD1-G93A)1Gur/J mouse strain have been sampled by laser microdissection. About 300 motor neurons were sampled per mouse (n = 7). For the laser microdissections 8 um coronal section of lumbar spinal cord was prepared and Leica Laser Microdissection system was used. These mice were treated with vehicle (250 mM sorbitol, 0.004% Tween-80 and 10 mM sodium acetate buffer). At this age mice presented clear motor impairment and motor neuron degeneration is documented.

Motor neurons from 11 weeks old mice of B6.Cg-Tg(SOD1-G93A)1Gur/J mouse strain have been sampled by laser microdissection. About 300 motor neurons were sampled per mouse (n = 8). For the laser microdissections 8 um coronal section of lumbar spinal cord was prepared and Leica Laser Microdissection system was used. At this age mice present no signs of motor dysfunction but subtle signs of denervation detectable by electromyography.

Liver obtained from wild-type male mice (CBA/J, 5 months old) fed AIN-93M-krill oil diet for 3 months. AIN-93M diet was modified by an equivalent amount substitution of the soy bean oil with 1.5% of krill oil. The content of docosahexaenoic acid (DHA) was 0.11 g per 100g of diet and the amount of eicosapentaenoic acid was 0.19 g per 100g of diet. Mice were euthanized by cervical dislocation after an overnight fast.
