Freshly isolated RhoA-null keratinocytes obtained from epidermis of RhoA (fl/fl) K5 Cre mice. Skin samples were washed for 5 min in PBS containing antibiotics followed by digestion with 0.8% trypsin in PBS (50 min; 37 °C). Obtained epidermis was shaken for 30 min in low-calcium MEM supplemented with 0.25 mg/ml DNAase I and 8 % FCS treated with Chelex. Cell suspension was filtered (70 um) and collected keratinocytes were washed once with DNase I medium, resuspended and analyzed. RhoA: Ras homolog gene family, member A.
Control RhoA knockout study 1 (in vitro)
Cultured primary RhoA-null keratinocytes obtained from epidermis of RhoA (fl/fl) K5 Cre mice. Primary keratinocytes were cultured in low-calcium MEM containing 5 ug/ml insulin, 10 ng/ml epidermal growth factor, 10 ug/ml transferrin, 10 uM phosphoethanolamine, 10 uM ethanolamine, 0.36 ug/ml hydrocortisone, 2 mM L-glutamine, 100 U/ml penicillin, 100 ug/ml streptomycin and 8% Chelex-treated FCS. CD8+ tumor infiltrating T-cell samples transduced with shRNA targeting Ppp2r2d were isolated from B16-OVA melanoma tumor which was developed in C57BL/6 mouse. IL-7/IL-15 cultured OT-I T cells were transduced with Ppp2r2d shRNA (pLKO.3G vector). Infected cells were sorted to purity using GFP encoded by the vector as a reporter. OT-I derived T cells transduced with shRNA were injected i.v. into mice bearing day 14 B16-Ova tumors. Seven days later, shRNA-expressing OT-I T cells (CD8+GFP+) were isolated from tumor. B16-Ova tumor cells were injected subcutaneously into C57BL/6 mice.

Lung samples obtained from M. tuberculosis H37Rv infected TNF-alpha -/- mouse strain on a mock (saline) treatment. One day before the infection with M. tuberculosis, mice (age at 8 - 12 weeks, female) were treated with saline. Subsequent saline treatment was applied in weekly intervals into infected mice. Pulmonary infection with M. tuberculosis H37Rv was performed under xylazine-ketamine anesthesia by delivering 20 ul of bacterial inoculum (1000±300 CFU) into each nostril. Twenty four hours post infection, the bacterial dose was confirmed in the lungs of 2 wild-type control mice. The experiment was terminated 1 month after M. tuberculosis inoculation and lung samples were processed for microarray analysis.
